Understanding next-generation sequencing

Short answer

Understanding next-generation sequencing means knowing how a laboratory reads many genetic changes at once to help guide diagnosis, treatment, or family planning.

This answers one question. It is not a diagnosis and does not replace an assessment by a licensed doctor.

At a glance

Sample
Blood, saliva, or tumour tissue may be used.
Scope
Many genes can be assessed in one test.
Interpretation
Results need clinical context.

What the test means

Next-generation sequencing examines DNA from a blood sample, saliva, or tumour tissue, depending on the clinical question. It can look at many genes in one test rather than examining each gene separately. In cancer care, tumour profiling may identify changes that help classify a tumour or suggest treatments. In inherited-condition testing, it may look for a variant that could be passed through a family.

A result may be positive, negative, or uncertain. A finding does not automatically explain every symptom, and a negative result cannot rule out every genetic cause. Interpretation depends on the sample, the genes included, the person's health history, and sometimes relatives' results.

What to do with a result

Ask which sample was tested, which genes or regions were included, and whether the result concerns the tumour or inherited DNA. Keep the laboratory report with your medical records. A genetic counsellor or specialist can explain whether a finding affects treatment, screening for relatives, reproductive decisions, or none of these. Do not change medicines or arrange family testing from a result label alone.

When to seek care

Contact the clinician who ordered the test when the report is difficult to understand, contains an uncertain finding, or recommends follow-up. Seek urgent medical help for new severe symptoms such as trouble breathing, confusion, heavy bleeding, or sudden weakness; the sequencing result itself does not determine how urgently a symptom needs assessment.

Questions for your doctor

What question was this sequencing test intended to answer? Does the result affect my diagnosis or treatment? Is the finding inherited or limited to the tumour? Should a genetic counsellor review it? Do relatives need advice or testing? Could the laboratory reclassify an uncertain result later?

Questions people ask

Is next-generation sequencing the same as a diagnosis?

No. It provides genetic information that clinicians interpret alongside symptoms, examination, imaging, and other tests.

Can a sequencing result affect family members?

It can, when a finding is inherited, so specialist counselling may be advised before relatives are tested.
