Estradiol is a form of oestrogen made mainly by the ovaries before menopause, with smaller contributions from other tissues. Levels naturally change across the menstrual cycle and are also influenced by pregnancy, menopause, hormone treatment, and some medicines. A result outside a laboratory's reference range is not a diagnosis by itself. Clinicians may use the test when assessing irregular periods, fertility concerns, symptoms related to menopause, or selected hormone conditions. They interpret it alongside the timing of the sample and other findings.

Before the appointment, tell the clinician about hormonal contraception, fertility medicines, gender-affirming hormones, supplements, and any recent treatment changes. Ask whether the sample should be taken at a particular point in your cycle and whether fasting is needed; do not assume either applies. The sample is collected from a vein and may leave mild, short-lived tenderness or bruising. Keep the result with the date and cycle or treatment details so it can be interpreted accurately.

Contact a clinician about unexpected bleeding, severe pelvic pain, fainting, or symptoms that are rapidly worsening, rather than waiting for the test result alone. Seek urgent help for heavy bleeding with weakness, collapse, or severe pain. If a result concerns you, arrange a review so the number is considered with your history; do not start, stop, or alter hormones based on a laboratory report without advice.
