Synaptophysin Immunohistochemistry

Synaptophysin is a protein associated with small vesicles in nerve and neuroendocrine cells. In immunohistochemistry, a laboratory antibody binds to the protein and creates a visible staining pattern under a microscope. A positive pattern can support a diagnosis involving neuroendocrine differentiation, but it is not a diagnosis by itself.

Pathologists interpret the distribution and strength of staining alongside the cell appearance, tissue architecture and other markers. A negative result does not answer every diagnostic question, and a positive result does not identify the tumour type without the rest of the pathology picture. Ask whether the report describes the staining as positive, negative, focal or diffuse, and whether the pattern is considered specific in your sample. The report may list additional stains used to distinguish neuroendocrine tumours from other cancers or from non-cancerous tissue.

The stain itself does not usually cause symptoms because it is performed on tissue already collected. Contact the treating team if the biopsy site becomes increasingly painful, red, swollen or draining, or if you develop fever. Seek prompt advice if you are unsure whether a reported result changes the next appointment or treatment plan.
