What should you know about F4/80 immunohistochemistry?

F4/80 is a marker associated with macrophages, a group of immune cells found in many tissues. In immunohistochemistry, laboratory staff apply an antibody to prepared tissue; the antibody binds to a matching target and creates a visible signal under a microscope. A pathologist then considers where the signal appears, how strong it is, and which cells show it.

A positive F4/80 stain does not by itself diagnose cancer, prove that a tumour is growing, or explain why an immune cell is present. Macrophages can occur in normal tissue, inflammation, healing tissue, and tumour environments. The result must be read with the tissue appearance, other stains, imaging, and clinical history. Human pathology reports may use related macrophage markers, so the exact wording and laboratory method matter.

Ask for the full pathology report and the specific sample location. Check whether the report describes F4/80 as positive, negative, focal, diffuse, weak, or strong, and whether a percentage or scoring method is used. Ask which other markers were tested and what diagnosis the complete panel supports. If the sample came from a biopsy, your treating team can explain how the finding fits with imaging and the next decision. Do not interpret one stain in isolation or change treatment based on it alone.

The stain itself does not cause symptoms because it is performed on tissue in the laboratory. Contact your clinical team promptly if the biopsy site develops spreading redness, increasing pain, pus, fever, or bleeding that does not settle with pressure. Ask when the pathology result is ready if uncertainty about the report is delaying an appointment or treatment discussion.
