The short answer

The stain is performed on biopsy or surgical tissue in the laboratory. A CK7-positive or CK7-negative result is a pattern clue, not a diagnosis by itself, because different tumour types can share the same staining result.

How CK7 is interpreted

Cytokeratins are structural proteins found in epithelial cells. The laboratory applies an antibody that binds to CK7 in preserved tissue, then the pathologist assesses where staining appears and how much of the sample shows it. The report may describe staining as positive, negative, focal, or diffuse.

Meaning comes from the combination of the cells' microscopic appearance, the CK7 pattern, other immunohistochemical markers, and the location of the sampled tissue. This panel approach can help narrow the likely tissue of origin or distinguish between possibilities that look similar under routine microscopy. CK7 alone cannot confirm where a cancer began or determine its stage.

What to do with the report

Review the complete pathology conclusion rather than reading the CK7 line in isolation. Check whether the sample was adequate, which other markers were tested, and whether the pathologist gives a definite diagnosis or a short list of possibilities. The clinician can connect that interpretation with scans, examination findings, and the site from which the tissue was collected.

If the wording remains uncertain, ask whether more stains, molecular testing, additional tissue, or review by a specialist pathologist would clarify the result. The appropriate next step depends on the whole diagnostic picture, not simply whether CK7 is positive or negative.

When to seek medical care

The CK7 stain itself is carried out in a laboratory and causes no new physical symptoms. Care advice instead relates to the biopsy or operation used to obtain the tissue. Follow the procedure-specific discharge guidance, and contact the treating team for worsening pain, increasing redness or swelling, fever, drainage, or bleeding that does not settle. Seek urgent help for severe bleeding, fainting, breathing difficulty, or another rapidly worsening symptom.

Useful questions include: Was CK7 positive throughout the sample or only in part? Which other markers shaped the pathologist's conclusion? Does the staining fit the suspected tissue of origin? Is the diagnosis final, or is further testing planned? Ask for the conclusion to be explained in plain language and for the next diagnostic or treatment decision it informs.

Does CK7-positive mean cancer?

No. CK7 expression can occur in different epithelial tissues, and the result must be interpreted with cell appearance, other stains, the tissue site, and clinical information.

What does CK7-negative mean?

It means the laboratory did not identify the expected CK7 staining pattern in the tested sample. That finding may help narrow diagnostic possibilities, but it does not exclude cancer or establish a tissue of origin on its own.

Why are other stains ordered with CK7?

Tumours can overlap in their CK7 pattern. Comparing several markers gives the pathologist a more informative profile and helps distinguish possibilities that may look alike under routine microscopy.
