Do I Need Amniocentesis After a Negative NIPT?

Amniocentesis is not automatically needed after a negative result. It may be offered when an ultrasound finding, family history, previous pregnancy, parental chromosome finding or another test raises a question that NIPT did not address. Your preference for greater diagnostic certainty also belongs in the decision.

NIPT analyses placental DNA fragments circulating in the pregnant person's blood. It estimates the chance of selected chromosome conditions and is therefore a screening test. Its scope depends on the panel used, and a low-chance result applies only to the conditions included.

Amniocentesis obtains amniotic fluid containing fetal cells. Those cells can undergo diagnostic chromosome or genetic analysis chosen for the clinical question. The procedure is invasive and carries possible complications, so the added information must be weighed against the procedural burden and your values.

Ask exactly which conditions your NIPT screened and whether the laboratory reported any limitation. Review ultrasound findings with your maternity or fetal medicine clinician. A genetic counsellor can explain which amniocentesis analysis would be ordered, what an unclear result could mean, and whether the result would change pregnancy care or preparation.

Consider how you feel about diagnostic uncertainty, an invasive procedure and the possibility of unexpected information. You can ask for time to discuss the choice with your support person, while also checking whether the proposed test has a pregnancy-stage window.

Contact your maternity team promptly if a scan has identified a concern or if you do not understand how the NIPT result relates to it. After amniocentesis, seek urgent advice for fluid leaking from the vagina, vaginal bleeding, fever, worsening abdominal pain or regular contractions. Follow the procedure team's specific discharge instructions.

Ask what prompted the offer of amniocentesis despite the negative NIPT, what the proposed laboratory analysis can and cannot detect, and how long results may take. Also clarify the procedure approach, possible complications, available alternatives and what follow-up each possible result would trigger.
