Next Generation Sequencing and PCR: What to Know

PCR, or polymerase chain reaction, makes many copies of a chosen DNA region. This can help a laboratory detect or enrich targets when the available material is limited. Next-generation sequencing then reads many DNA fragments in parallel, depending on the design of the test. Some workflows use PCR before sequencing, while others use different preparation methods.

The combination does not mean every gene or every change has been examined. PCR-based enrichment focuses attention on selected regions, and the result depends on the sample, laboratory method, and quality of the material. In oncology, such testing may be part of genomic tumour profiling. Similar laboratory principles can support inherited testing, carrier screening, or other defined genetic questions.

Ask which DNA regions are targeted, whether the test is designed to find a known change or scan a broader panel, and whether the sample is blood or tumour tissue. Confirm whether a positive finding needs another method for verification. Ask how low-quality or insufficient material is handled and who will discuss findings that could have family implications.

Testing itself does not usually explain new symptoms. Contact your clinical team when a report recommends urgent assessment, repeat sampling, or a treatment discussion. Seek emergency care for severe breathing difficulty, sudden weakness, uncontrolled bleeding, fainting, or rapidly worsening pain; do not wait for PCR or sequencing results in that situation.
