The short answer

The stain shows whether sampled cells contain certain cytokeratin proteins. Its pattern is interpreted with the cell appearance, tissue architecture and other stains before the pathologist reaches a diagnosis.

Reading CK5/6 in context

DCIS means abnormal cancer cells are contained within breast ducts, without invasion into surrounding breast tissue. CK5/6 staining can help separate some usual, mixed-cell duct changes from more uniform clonal proliferations. A patchwork pattern and an absent pattern can carry different diagnostic clues, but neither result is universal for every DCIS subtype. Positive staining does not mean that disease has spread, and negative staining does not by itself prove DCIS.

Using the result in care decisions

Read the CK5/6 line alongside the final diagnosis, grade, size or extent, margin status and hormone-receptor findings. These features, imaging and personal health priorities shape the care discussion. Depending on the complete picture, local treatment may involve lumpectomy, radiotherapy or mastectomy; hormone therapy may be considered when the DCIS has the relevant receptors. If the report says the stain is equivocal, limited or part of a differential diagnosis, ask whether deeper sections, additional markers or specialist pathology review are needed.

When to contact the care team

A CK5/6 result itself does not create an emergency. After a breast biopsy or operation, contact the clinical team promptly for increasing redness, warmth, drainage, fever, worsening pain or swelling that continues to enlarge. Seek urgent care for bleeding that does not stop with firm pressure, rapidly expanding breast swelling, faintness or breathing difficulty. These concerns relate to recovery from the procedure and require assessment regardless of the stain result.

Questions that clarify the pathology report

Ask what diagnostic question the CK5/6 stain was intended to answer and whether its pattern changed the final interpretation. Request a plain-language explanation of the DCIS grade, extent, margins and receptor status. If surgery is being planned, clarify whether imaging and biopsy findings agree, what tissue will be removed, and which pathology results could alter the next treatment discussion.

Questions people ask

Does CK5/6-positive DCIS mean invasive breast cancer?

No. CK5/6 describes protein staining in cells. Invasion is decided by the tissue architecture and whether abnormal cells have crossed the duct boundary, not by CK5/6 positivity alone.

Does a negative CK5/6 stain confirm DCIS?

No. An absent stain may support a pathologist's interpretation in a particular pattern, but the diagnosis relies on the tissue appearance and the combined findings.

Is CK5/6 the same as a hormone-receptor test?

No. CK5/6 detects cytokeratin proteins. Hormone-receptor tests answer a different question and can influence whether hormone therapy is discussed.
