Why are PARP inhibitor drugs used in PARP inhibitor therapy?

PARP proteins help repair certain types of damage in DNA. Some tumours already have weakness in another repair pathway, sometimes involving BRCA-related or other repair genes. When PARP is blocked, the tumour cell may struggle to cope with accumulated DNA damage and stop growing or die. This effect is selective in intent, but healthy cells can also be affected.

Whether treatment is suitable depends on the cancer type, previous therapy, tumour or inherited test results, blood counts, organ function, and local treatment guidance. A repair-gene change does not guarantee benefit, and absence of a recognised change does not make every treatment decision simple.

Ask how the medicine will be taken, what monitoring is planned, and whether a tumour test or inherited cancer test is relevant. Keep appointments for blood tests because these medicines can lower red cells, white cells, or platelets. Report new breathlessness, unusual bruising, fever, persistent nausea, or marked fatigue rather than adjusting the medicine yourself.

Contact your treatment team promptly for fever, signs of infection, unusual bleeding, severe weakness, worsening shortness of breath, chest discomfort, or persistent vomiting. Emergency help is appropriate for severe breathing difficulty, collapse, or confusion. The team may pause, adjust, or change treatment after checking the cause.
