What should you know about retinoblastoma immunohistochemistry?

What the laboratory test does

A pathologist prepares a thin section of tissue and applies antibodies designed to bind to selected proteins. A chemical or fluorescent label makes the binding pattern visible under a microscope. The result is interpreted alongside the appearance of the cells, the child’s examination and imaging findings, rather than as a stand-alone answer.

In suspected retinoblastoma, this testing can help compare the sample with other small round cell tumours that may look alike under routine staining. A positive or negative stain does not by itself describe how a tumour will behave, and weak or mixed staining may need careful interpretation.

What happens next

Ask whether the sample contains enough viable tissue for the planned tests and whether a specialist eye pathologist will review it. The report may list the markers tested, the staining pattern and the pathologist’s overall interpretation. Your child’s oncology team can explain how those findings fit with scans, examination and any genetic testing.

If tissue has not yet been obtained, the team will discuss whether a biopsy is appropriate. In retinoblastoma, reaching a diagnosis safely is especially important because procedures involving the eye may carry risks to vision and require specialist planning.

When to seek urgent care

Contact the treating team promptly for a sudden change in vision, increasing eye redness or swelling, severe eye pain, repeated vomiting, marked sleepiness, or a fever after a procedure. Follow the discharge instructions for wound care and medicines, and do not wait for a pathology appointment if a child appears acutely unwell.
