What should you know about PAX8 immunohistochemistry?

Immunohistochemistry uses antibodies to highlight selected proteins in cells. PAX8 is a nuclear marker that can support a pathologist’s assessment of whether a tumour may have arisen from tissues such as the kidney, thyroid, or Müllerian tract. A positive stain is a clue, not a diagnosis. Some tumours from other sites can show staining, and some relevant tumours may show little or no staining.

The pathologist reads PAX8 alongside the cell pattern, tumour location, imaging, clinical history, and a panel of other stains. The wording may describe staining as positive, negative, weak, focal, or diffuse. Those terms describe the laboratory finding; they do not independently state how serious the cancer is or which treatment is needed.

Ask for the complete pathology report and have the treating clinician explain how PAX8 fits with the biopsy diagnosis. Ask which other stains were used and whether the panel supports a likely tissue of origin. If the sample is small or the result is unclear, the team may request additional testing, review the slides, or compare the finding with scans. Treatment decisions should follow the integrated diagnosis rather than one stain in isolation.

PAX8 staining itself does not cause symptoms and is not an emergency. Seek prompt clinical advice if you have new bleeding, severe pain, rapidly worsening swelling, difficulty breathing, or a sudden change in your general condition while waiting for results. For report questions, contact the clinician who arranged the biopsy so that an unexplained result is not interpreted without its clinical context.
