What should you know about JC-1 flow cytometry?

How the test works

JC-1 is a dye whose fluorescence changes according to the electrical state across a cell’s mitochondrial membrane. In flow cytometry, cells pass one by one through a laser-based instrument. The machine records fluorescence patterns from many cells, allowing the laboratory to compare populations with different mitochondrial signals.

The method can support laboratory work on cell health, programmed cell death, or treatment response, depending on the sample and the question being investigated. A JC-1 result is not a diagnosis on its own. Its meaning depends on controls, sample quality, the cell type, and the laboratory’s interpretation.

Preparing for a result

Ask which sample is being tested and what clinical or laboratory question the test is intended to answer. If the sample comes from a bone marrow biopsy or another procedure, follow the preparation and aftercare instructions for that procedure. Keep the report with other pathology results, and ask whether the finding will be combined with microscopy, immunophenotyping, genetic testing, or other measurements.

When to contact the care team

The laboratory test itself does not usually create symptoms that require emergency care, but the sample collection procedure may have aftercare warnings. Contact the team for fever, increasing redness or drainage, heavy bleeding, severe pain, or feeling faint after a biopsy or blood draw. Ask promptly if a result is unclear, unexpected, or reported without the clinical context needed to understand it.
