What should you know about intracellular staining flow cytometry?

A laboratory receives cells from blood, bone marrow or another sample. The cells are treated to preserve their identity and make their outer membrane permeable. Special antibodies can then enter the cells and attach to specific internal proteins. Fluorescent labels on those antibodies help the flow cytometer recognise the signals.

The instrument examines cells one by one and records several features at once, including cell size, granularity and marker patterns. A specialist interprets the combined pattern rather than relying on one signal alone. The result may help classify abnormal cells, assess cell activity or support a diagnosis alongside examination, imaging and other laboratory findings.

This is mainly a laboratory process, so your experience depends on how the sample is collected. A blood sample usually involves a brief needle prick. A bone marrow sample is collected by a separate procedure and may cause temporary soreness. Ask whether you need to avoid food or change medicines; preparation varies by the sample and clinical question.

Results can take time because the laboratory must process, stain and analyse the cells. The report may describe which markers were detected and how they were distributed across cell groups. Your clinician combines that information with your symptoms, examination and other test results.

Follow the collection team’s wound-care instructions. Seek advice for bleeding that does not settle, increasing redness, drainage, severe pain or fever after an invasive sample. A small bruise or short-lived tenderness can occur after a needle-based collection.

Ask which cell markers are being examined, what sample is required, when the report is likely to be ready, and how the findings fit with your other tests. Ask whether a repeat sample or additional testing could be needed if the cell pattern is unclear.
